Approach from Berlin
Take the A24 in direction on Hamburg to the Wittstock/Doss-intersection, further on the A19 to the Rostock-cross, then the A20 to Stralsund, the B96 over the Rügendamm to the Isle of Rügen, then go on far as Lietzow.
or:
Take the B96 via Neubrandenburg, Greifswald and Stralsund, over the Rügendamm to the Isle of Rügen and further in direction on Sassnitz to Lietzow.
Approach from Hamburg
Take the A1 till aou reach the Lübeck-intersection, the A20 to Stralsund, the B96 over the Rügendamm, further as seen above.
